Introduction Infection surveillance methods in hospitals are of great concern because of the high frequency of the disease. Diagnostic techniques, such as polymerase chain reaction (PCR) assay and reverse transcriptase-polymerase chain reaction (RT-PCR), are applied in clinical studies for the detection of the infection in hospitalized patients. Many infectious diseases cause mortality and morbidity in humans; therefore, methods for early detection of the infection become increasingly important. Currently, there are many methods available for the analysis and detection of infectious diseases, such as conventional serological methods, immunoblots, enzyme linked immunosorbent assays (ELISA), latex agglutination tests, and immunofluorescence assays (IFA). These methods are applied in clinical studies because of their low-cost and sensitivity. For example, RT-PCR is a simple, rapid, and cost-effective method to detect the infection in clinical studies. However, PCR can only analyze the microorganisms associated with the infection. Currently, PCR is often used to detect the presence of the *Mycobacterium tuberculosis complex* or *Mycobacterium avium* in clinical studies. However, such tests require special equipment, such as centrifuges, special instruments, a thermal cycler, and gel electrophoresis. Moreover, these tests are also very expensive. In the past few years, there have been some improvements in the sensitivity of PCR. For example, the technique can detect less than 2 colonies/reaction in RT-PCR. PCR is also a very sensitive technique and does not require any special instruments for diagnosis. The current PCR is used in clinical studies to identify the *Mycobacterium tuberculosis* complex in patients or control patients. In addition, because of its fast detection speed through the amplification of genomic DNA, PCR is also widely applied to detect other infectious diseases. For example, RT-PCR can detect the pathogen in less than 10 min after the onset of symptoms. The PCR is also applicable to many other infectious diseases, such as brucellosis, tick-borne encephalitis, Lyme disease, dengue fever, and hepatitis B. In summary, RT-PCR is an effective and inexpensive method for the detection of both viral and bacterial infections.
