The implications of this work for the development of potential therapies for human neuropsychiatric disorders and for the application of this information to the development of a therapeutic that can enhance the activity of neural circuits as well as restore or normalize cognitive processes and behavior are discussed in this article. Neuropsychiatric illnesses such as schizophrenia, psychosis, and depression share common pathologic cellular and network abnormalities that may be amenable to targeted treatment with compounds that act on neural networks in ways that have yet to be explored in animals. This article will be of interest to scientists who study molecular and cellular neurobiology, neurobiology, computational neurobiology, and computational neuroscience (see below). The authors were led by Dr. Alan Kahan, a postdoctoral researcher at the National Institute on Drug Abuse (NIDA), and are Michael V. Cohen, associate professor, graduate student, and William G. Cissell, professor, and assistant research scientist, and are collaborators on this work with Dr. Kalevsky, professor, and director of NIDA’s Neurobehavioral Research Center. The paper titled “Translational neuroscience: Neuronal circuits are affected by a single dose of psilocybin” was published in the October 25, 2018 issue of Journal of Neuroscience.

The paper presents the results of two experiments investigating the effects of the psilocybin-containing ingredient of magic mushrooms. The first experiment measured the effects of a single dose of psilocybin in multiple areas of the brain on the behavior of a rat. The results reveal that a single dose of psilocybin has a significantly greater effect in the same location of the brain that is involved in producing a natural antidepressant effect, as opposed to that involved in producing a natural mood elevation effect.

Abstract: The authors studied the effects of ketamine (Ket) and psilocybin (Psi) on the morphology, dendritic spine density, and activity of hippocampal neurons in the rat. Hippocampal slices were prepared from adult rats, which were divided into four groups: saline + control group (SA), saline + ketamine (Ket) group (SKA), saline + ketamine + Psi group (SKA+Psi), and ketamine + Psi group (KA+Psi). Neurons were cultured for 36 hours with KA+Psi to determine the effects of ketamine and Psi on the morphological change of dendritic spines. These effects were assessed by a morphological assay, a quantitative assay, and an activity test. There was no difference in the dendritic spine density between the saline + control and saline + Psi groups, whereas the dendritic spine density in the SA and SKA groups was significantly decreased. The morphological analysis revealed that the dendritic spines of neurons in the KA, KA+Psi, and SA groups were significantly fewer than in the SA group. The morphological effect was more obvious in the KA+Psi group. There was no difference in the number of dendritic spines between the SA and SKA groups. In the SA and SKA+Psi groups, KA+Psi treatment significantly improved learning abilities, memory, and memory consolidation. These effects were more obvious in the SA and SKA+Psi groups. The dendritic spine density in the KA+Psi group was significantly increased. These results suggested that psilocybin can improve the morphology of the dendritic spines and enhance memory abilities in rats.
